    Autophagy inhibitors hydroxychloroquine

Note: based on a RGCC chemosensitivity analysis I have seen at a German clinic, Hydroxychloroquine has been effective in killing the cancer cells of 5 out 7 patients that were tested. It is one of very few available drugs that inhibits autophagy, a mechanism associated with its anticancer properties.

The combination of temsirolimus TEM, an MTOR inhibitor, and hydroxychloroquine HCQ, an autophagy inhibitor, augments cell death in preclinical models. This phase 1 dose-escalation study evaluat. Hydroxychloroquine Sulfate is an antimalarial agent used for the treatment of systemic lupus erythematosus, rheumatoid arthritis and other autoimmune, inflammatory and dermatologic conditions. Also acts as an inhibitor of autophagy and toll-like receptor TLR 7/9. Hydroxychloroquine PLAQUENILR is an orally available autophagy inhibitor that has been used for many years to treat autoimmune diseases like lupus, sarcoidosis and rheumatoid arthritis at the standard dosing of 400-600mg/day.

In cancer, authophagy is the process used by cancer cells to “self-eat” in order to survive. Specifically, if authophagy is prolonged this will become a lethal process to cancer. duing chemotheraphy, radiotheraphy, etc.) authophagy is used by cancer cells to survive. However, note that Chloroquine has other properties as well that may be very well related to anti cancer mechanism, such as zinc ionophore, and others (see below the section on mechanisms).

  7. Tumor cells require autophagy to remove misfolded proteins or damaged cellular organelles. SBI-0206965 was shown to suppress autophagy induced by mTOR inhibition. mTOR signaling is often hyperactive in many tumors and can drive proliferation, so mTOR inhibitors are being investigated as anticancer agents.

As shown in preclinical models, inhibition of autophagy restored chemosensitivity and enhanced tumor cell death. These results established autophagy as a therapeutic target and led to multiple early phase clinical trials in humans to evaluate autophagy inhibition using hydroxychloroquine in combination with chemotherapy or targeted agents. Autophagy inhibition remains a very attractive strategy to augment standard therapy and improve long-term outcomes for a broad range of malignancies. Our collective work, to date, has demonstrated that we are finally on a strategic trajectory to move beyond HCQ with respect to clinical autophagy inhibition. Animal and cellular studies support the importance of autophagy inhibition in lymphangioleiomyomatosis LAM. In a cohort of subjects with LAM, we tested the hypothesis that treatment with sirolimus and hydroxychloroquine an autophagy inhibitor at two different dose levels is safe and well tolerated. Secondary end points included changes in lung function.

500 mg chloroquine phosphate (300 mg base) orally on the same day each week Comments: -If possible, suppressive therapy should start 2 weeks prior to exposure; if unable to start 2 weeks before exposure, an initial loading dose of 1 g chloroquine phosphate (600 mg base) may be taken orally in 2 divided doses, 6 hours apart. Plaquenil and Prednisone By Connie Rifenburg June 3, 2015 at pm. Ang, I had hair loss pretty severe when I first started on Prednisone, Plaquenil, and Methotrexate. I guess it may depend on the dosage, and hopefully your hair loss will stop as mine did after a few months.

The following adverse reactions have been identified during post-approval use of PLAQUENIL or other 4-aminoqunoline compounds. Because these reactions are reported voluntarily from a population of uncertain size, it is not always possible to reliably estimate their frequency or establish a causal relationship to drug exposure.
