Plaquenil aps Chloroquine for malaria prevention

Jan 01, 2020 Plaquenil® hydroxychloroquine sulfate is a white or practically white, crystalline powder, freely soluble in water; practically insoluble in alcohol, chloroform, and in ether. The chemical name for hydroxychloroquine sulfate is 2-4-7-Chloro-4-quinolylaminopentylethylamino ethanol sulfate 11. Hydroxychloroquine Plaquenil is also a disease-modifying anti-rheumatic drug DMARD used to treat both acute and chronic rheumatoid arthritis, systemic lupus erythematosus, and discoid lupus erythematosus. Hydroxychloroquine likely works in the prevention and treatment of malaria by interfering with the parasite's normal growth and replication process. Irreversible retinal damage observed in some patients who had received hydroxychloroquine sulfate; significant risk factors for retinal damage include daily doses of hydroxychloroquine sulfate greater than 6.5 mg/kg 5 mg/kg base of actual body weight, durations of use greater than five years, subnormal glomerular filtration, use of some concomitant drug products such as tamoxifen citrate and concurrent macular disease

Today, it is used to treat rheumatoid arthritis, some symptoms of lupus, childhood arthritis (or juvenile idiopathic arthritis) and other autoimmune diseases. Hydroxychloroquine is in a class of medications that was first used to prevent and treat malaria.

Hydroxychloroquine is an aminoquinoline like chloroquine. It is a commonly prescribed medication in the treatment of uncomplicated malaria, rheumatoid arthritis, chronic discoid lupus erythematosus, and systemic lupus erythematosus. Hydroxychloroquine is also used for the prophylaxis of malaria in regions where chloroquine resistance is unlikely. Originally, hydroxychloroquine was used to prevent and treat malaria and was considered very effective. Now it is generally used for rheumatoid arthritis, but it has also been shown to work well for juvenile arthritis, some lupus symptoms, and other types of autoimmune conditions. Mechanism Hydroxychloroquine is thought to improve symptoms of systemic diseases by preventing inflammation. For example, to treat both rheumatoid arthritis and systemic lupus erythematosus SLE, it has been hypothesized that the drug suppresses T-lymphocytes by increasing the pH inside lysosomes, which disables leukocyte chemotaxis. This prevents leukocytes from being activated which decrease the inflammation seen in many systemic diseases.
